Abstract
Geranyl-modified oligonucleotides can anchor onto neutral membranes and oil droplets.
- The development of an efficient method for modifying oligonucleotides with geranyl groups in water was achieved.
- Geranyl-modification can be replaced with thiol-nucleophiles, allowing for a reversible change in the oligonucleotides' hydrophobic properties.
- Modified oligonucleotides attach to neutral membrane structures, potentially enhancing delivery systems.
- Oligonucleotides on the surface of neutral liposomes can move to the inner membrane surface through freeze-thaw cycles, which may protect them from degradation.
- These findings could provide new strategies for oligonucleotide lipidation in RNA delivery.
