TRPML3 and TRPML1/3 differently control unusual MIF release through TMED10 and secretory autophagy

Updated

Abstract

TRPML3 regulates type III unconventional protein secretion (UcPS) through two distinct pathways.

  • TRPML3 interacts with TMED10 and macrophage migration inhibitory factor (MIF), facilitating MIF secretion.
  • Cytosolic MIF is transported to membrane-enclosed compartments via TMED10-channeled UcPS.
  • TRPML3 promotes TMED10 oligomerization, enhancing MIF secretion in a Ca2+-independent manner.
  • MIF colocalizes with LC3 in vesicular compartments, with its secretion dependent on secretory autophagy.
  • PI(3,4)P2 is identified as a key regulator of secretory autophagy, with TRPML1/3 mediating Ca2+-dependent MIF secretion.
