IL-1β (5 ng/mL) successfully induced a staged rat chondrocyte senescence model with defined phenotypes at 12, 24, and 48 hours.

  • Cell viability and cell-cycle analysis confirmed that prolonged IL-1β exposure led to an increase in senescence markers and mitochondrial damage.
  • The proportion of senescence-associated β-galactosidase-positive cells increased significantly with prolonged IL-1β induction.
  • Mitochondrial membrane potential declined progressively, correlating with the stages of chondrocyte senescence.
  • Upregulation of senescence markers and catabolic genes was observed, while anabolic gene expression decreased during senescence.
  • TSPAN9 overexpression reversed pathological changes associated with senescence, improved mitochondrial function, and restored ECM balance.
