An mRNA vaccine using a tagged E2 protein fully protects pigs from deadly classical swine fever virus

Abstract

Immunization with Ub-E2-mRNA-LNP provided complete protection against the highly virulent CSFV Shimen strain in piglets.

  • Rabbits immunized with XCL1-E2-mRNA-LNP or Ub-E2-mRNA-LNP showed no fever or detectable viral RNA in their spleens after challenge.
  • Ub-E2-mRNA-LNP induced stronger antibody and cell-mediated immune responses compared to other vaccine candidates.
  • Both XCL1-E2-mRNA-LNP and Ub-E2-mRNA-LNP offered strong protection against the live attenuated CSFV C-strain in rabbits.
  • The Ub-E2-mRNA-LNP vaccine outperformed the traditional E2-based subunit vaccine and other mRNA-LNP formulations.
