Urolithin A may protect kidneys from calcium oxalate crystals and damage by controlling PCK1 to restore mitochondrial cleanup in kidney stones

Updated

Abstract

Urolithin A (UA) markedly reduced renal calcium oxalate deposition and improved renal function in mice.

  • UA alleviated kidney injury associated with calcium oxalate crystals.
  • Both in vivo and in vitro studies showed that UA restored mitophagy suppressed by oxalate.
  • UA reduced oxidative stress and apoptosis linked to mitochondrial dysfunction.
  • Phosphoenolpyruvate carboxykinase 1 (PCK1) was identified as a key target of UA.
  • Inhibition of PCK1 diminished UA's protective effects, while its overexpression enhanced mitophagy under oxalate stress.
