Urolithin D Improves Thinking and Behavior in Aging Mice by Helping Cells Clear Damaged Mitochondria Through Energy and Survival Pathways

Updated

Abstract

Daily treatment with 50-150 mg/kg Urolithin D over 8 weeks improved cognitive performance and reduced hippocampal pathology in a D-galactose-induced aging mouse model.

  • Urolithin D treatment preserved synaptic plasticity and alleviated behavioral impairments associated with aging.
  • Increased expression of proteins related to mitochondrial function, such as AMPK, PGC-1α, and SIRT3, was observed with Urolithin D administration.
  • Urolithin D influenced mitophagy-related signaling by altering the expression of proteins like Beclin1, LC3, and Parkin.
  • In vitro experiments indicated that Urolithin D enhanced mitochondrial integrity and reduced oxidative stress in neuronal cells under stress.
  • Knockdown of SIRT3 in HT22 cells led to increased mitochondrial damage and reduced activation of mitophagy-related processes.
