Impaired pexophagy leads to marked accumulation of the peroxisomal membrane protein PMP70 in renal tubular epithelial cells under diabetic conditions.

  • Diabetic kidney disease is associated with progressive tubular injury and peroxisomal dysfunction.
  • High-glucose conditions lead to significant elevation of USP30 in renal tubular epithelial cells.
  • Global genetic depletion of USP30 restores peroxisomal function and enhances pexophagy.
  • USP30 suppresses pexophagy by antagonizing the ubiquitination of the peroxisomal import receptor PEX5.
  • Deletion of the autophagy factor ATG5 negates the protective effects of USP30 deficiency.
