Human 3D Brain Model with Blood Flow on a Chip

Abstract

A 3D Brain-Chip system was developed incorporating multiple human-derived brain cell types to model the blood-brain barrier.

  • The system includes endothelial cells, pericytes, astrocytes, neurons, microglia, and oligodendroglia derived from induced pluripotent stem cells.
  • It allows for the modeling of brain microvasculature associated with sporadic Alzheimer's disease.
  • The platform enables systematic assessment of blood-brain barrier vascular morphology and function.
  • It is designed for real-time monitoring and screening of therapeutics aimed at optimizing delivery to the brain.
  • This multicellular 3D co-culture system may address limitations of existing brain-on-chip models by capturing the full cellular complexity of the human neurovascular unit.
