Overactive immune sensor in artery muscle cells worsens artery wall thickening in mice

Updated

Abstract

Vascular smooth muscle cell (VSMC) NLRP3 gain-of-function was associated with significantly exacerbated neointimal lesion growth in a mouse model.

  • Increased caspase1 activation was observed in VSMC-specific Nlrp3 gain-of-function mice, both in vivo and after carotid partial ligation.
  • VSMC Nlrp3 gain-of-function led to heightened vascular inflammation, including increased VCAM1 expression and macrophage accumulation.
  • Enhanced Gasdermin D signaling and increased TUNEL-positive cell death were noted in the arteries of Nlrp3 gain-of-function mice.
  • The model exhibited greater intimal area and intima-to-media ratio, indicating more severe neointimal lesion growth.
  • Lipid accumulation and foam cell-like remodeling were observed in the injured arterial wall, along with reduced expression of the transcription factor EB.
