Abstract

BCL2 was identified as a key regulator of tumor growth after radiation in diffuse midline gliomas (DMGs).

  • Radiation sensitizes DMGs to the BCL2 inhibitor venetoclax, regardless of p53 status.
  • Venetoclax alone did not cause significant cell death in DMG cells.
  • Post-radiation treatment with venetoclax significantly increased cell death and apoptosis.
  • Venetoclax treatment reduced the association between BCL2 and BH3 apoptosis initiators.
  • Combining venetoclax with radiotherapy significantly enhanced survival in mice with DMG tumors.
