Abstract
The optimized ATAC-see platform enables high-throughput quantitative imaging of chromatin states in fixed mammalian cells and fresh frozen tissues.
- Progressive age-associated chromatin opening and heterochromatin remodeling were observed in primary human fibroblasts.
- Rapid, reversible chromatin reorganization occurred during partial reprogramming of aged fibroblasts driven by OSK(M).
- The protocol was successfully adapted to murine tissue sections, facilitating in situ quantification of age-dependent remodeling.
- This framework positions chromatin accessibility as a scalable imaging biomarker for further studies on aging and rejuvenation.
