Brain white matter differences connect irregular daily activity patterns to mania symptoms in people at risk for bipolar disorder

Participants aged 16-24 (N = 112) showed that lower circadian rhythm robustness was linked to greater white matter disorganization in specific brain pathways.

  • At higher levels of mania vulnerability, greater instability in sleep patterns was associated with increased white matter disorganization in the cingulum bundle and uncinate fasciculus.
  • Lower circadian function index scores correlated with higher orientation dispersion index values, indicating disrupted white matter organization.
  • Greater white matter disorganization in the uncinate fasciculus predicted more severe mania symptoms at a 6-month follow-up.
  • Findings suggest that instability in rest-activity rhythms may contribute to white matter disruption, potentially increasing the risk of mania in vulnerable individuals.
