Design of a wireless probe for controlled gene delivery using electrical pulses

Updated

Abstract

The novel gene electrotransfer probe may expand the transfection zone by up to 3× in cell studies.

  • A disposable needle-type probe was developed for precise delivery of DNA/mRNA to deep tissues using focused electric fields.
  • The probe utilizes a ganged linear bionic electrode array enhanced by integrated microfluidics to deliver DNA expression cassettes.
  • This system includes a wireless design powered by an integrated capacitor with an automated discharge mechanism.
  • Validation in cell studies showed that the conductivity-clamping method significantly increased the area of successful gene delivery.
  • In mouse models, the probe demonstrated proof of concept for deep tissue gene delivery with sustained expression of a reporter gene.
