A New Drug May Increase Radioactive Iodine Absorption by Boosting Natural Iodine Transport in Aggressive Thyroid Cancer

Abstract

CTOM-DHP, a newly identified small-molecule tyrosine kinase inhibitor, enhances NIS expression in anaplastic thyroid cancer cells.

  • Increased NIS expression was confirmed through promoter assays and mRNA/protein analyses after treatment with CTOM-DHP.
  • CTOM-DHP treatment led to higher expression of thyroid-specific proteins and transcription factors related to iodide metabolism.
  • NIS function was augmented, demonstrated by increased iodide uptake and cytotoxic effects.
  • The mechanism of action may involve the inhibition of PI3K/Akt and MAPK signaling pathways.
  • In vivo results indicated improved RAI avidity in a tumor xenograft mouse model after CTOM-DHP treatment.
