Fast CRISPR-based test for detecting grapevine black-foot disease pathogens

Abstract

The assay detects Ilyonectria DNA at concentrations as low as 3.6 ng/µL within 1 hour at 39 °C.

  • A nucleic acid assay combining recombinase polymerase amplification and CRISPR-Cas12a was developed for detecting grapevine black-foot disease pathogens.
  • The assay showed a detection limit of 80 fg/µL for Dactylonectria within 50 minutes at 41 °C.
  • No cross-reactivity was observed with non-target fungi during testing.
  • The lateral flow assay provided rapid visual results, allowing for quick interpretation.
