Targeted gene editors effectively fix PLP1 mutations in brain cells that make myelin

Spatially concentrating adenine base-editor activity improved correction of PLP1 mutations in hard-to-edit oligodendrocytes.

Evidence

A preclinical base-editing platform study used SunTag recruitment and compact Cas9 variants to test cABE editing of PLP1A243V in oligodendrocytes and measured localization, RNA off-target effects, and myelination-related phenotypes.

Caveat

The findings support a technical framework for PMD gene therapy, but human delivery, durability, and clinical benefit were not tested.
