Abstract

ALKBH5 downregulation significantly mitigated lung tissue damage and reduced pro-inflammatory cytokines in an LPS-induced murine ARDS model.

  • ALKBH5 regulates mA methylation in alveolar macrophages.
  • LPS stimulation decreases mA modification of ULK1 mRNA, which can be reversed by ALKBH5 downregulation.
  • Knockdown of ALKBH5 significantly suppresses autophagy and inhibits M1 pro-inflammatory polarization in macrophages.
  • In vivo, targeting ALKBH5 resulted in reduced pulmonary edema and decreased levels of IL-1β, TNF-α, and IL-17.
