Boosting Cas12a Activity by Changing DNA Shape for Fast and Direct Detection of DNA-Cutting Enzymes

A 1.8 Å increase in distance between Lys595 of Cas12a and the PAM may enhance binding free energy by 24.2 kcal/mol.

  • An allosteric Y-shaped DNA structure was developed to activate Cas12a through engineered PAM topologies.
  • This design features a split PAM at a three-way junction, which initially inhibits Cas12a activation.
  • Cleavage of the target releases fragments that can correct the PAM, restoring Cas12a's active conformation.
