Abstract

GL9, a guanidine-based lipid, enhances mRNA expression in lipid nanoparticles by 12-fold in the spleen after intraperitoneal injection.

  • The library of guanidine-based lipids was screened to improve mRNA delivery via lipid nanoparticles.
  • GL9 was identified as the optimal lipid for incorporation into lipid nanoparticles at a 5% molar ratio.
  • Transfection improvements were observed across various cell types and lipid nanoparticle formulations with GL9.
  • Incorporation of GL9 did not significantly change the physicochemical properties of the lipid nanoparticles.
  • GL9 increased mRNA expression by 2.7-fold following intramuscular administration.
  • GL9 also promoted dendritic cell maturation, indicating its potential role as an adjuvant in mRNA vaccines.
