New Amylin-Based Treatments for Weight Loss in Adults with Overweight or Obesity Without Diabetes: A Comparative Analysis

Abstract

In a meta-analysis of six trials involving 4642 participants, high dose subcutaneous amycretin resulted in a mean weight reduction of -23.95% compared to placebo.

  • High dose amycretin, eloralintide, and CagriSema were associated with the largest reductions in body weight among long-acting amylin-based therapies.
  • Other treatments such as semaglutide and liraglutide resulted in smaller weight reductions of -11.45% and -6.4%, respectively.
  • Similar trends were observed for reductions in body mass index and waist circumference.
  • Gastrointestinal adverse events, including nausea, vomiting, and constipation, were more prevalent with high dose amylin-based therapies.
  • Only high dose CagriSema led to an increase in treatment discontinuation due to adverse events.
  • Findings are preliminary and require confirmation in larger, more definitive trials.
