Semaglutide slowed aging and extended lifespan in older female mice
GLP-1 drugs had a big week, and not just for waistlines.
From mouse longevity to cancer risk reduction to sleep apnea, the evidence keeps expanding—and so do the complications worth watching.
🐭 A Weight-Loss Drug Just Extended Mouse Lifespan
- Researchers treated 20-month-old female mice (roughly equivalent to late middle age) with semaglutide for 3 months. The drug improved physical function, dialed back biological markers of aging, and—when continued—extended lifespan.
- The effects closely mirrored calorie restriction, a well-studied longevity intervention. In direct comparison, semaglutide matched or outperformed calorie restriction on spatial memory, exploratory drive, and glucose control.
- This is mice, not humans, and only female mice at that. But the mechanistic overlap with calorie restriction gives researchers a framework for why GLP-1 drugs produce benefits that go well beyond appetite suppression.
Why it matters: It reframes semaglutide from a metabolic drug into something that may interact with core aging biology—a much bigger claim that now needs human investigation.
Key Findings
💊 Tirzepatide vs. Semaglutide: The Head-to-Head Numbers
- Across 10 studies and 41,381 participants, tirzepatide produced about 4.3 percentage points more weight loss than semaglutide, plus an extra 4.4 kg in absolute terms.
- The catch: serious adverse events were 83% more frequent with tirzepatide. Gastrointestinal side effects were similar between the two drugs.
🔄 What Happens When You Stop
- A Bayesian meta-analysis reconstructed weight regain trajectories after stopping semaglutide or tirzepatide. The pattern is consistent: substantial weight returns after discontinuation, though the exact rate and ceiling remain uncertain.
- This reinforces that these drugs work while you take them—not as a one-time reset.
🎗️ GLP-1 Drugs and Cancer Risk: A Large Signal
- A meta-analysis of 78 observational studies found GLP-1 receptor agonist use was associated with statistically significant reductions in 10 of 13 obesity-linked cancers, including colorectal, liver, and pancreatic cancers.
- Compared specifically to insulin, the risk reductions were striking—colorectal cancer risk was nearly halved. Thyroid cancer showed a slight uptick, but it didn't reach statistical significance.
😴 Sleep Apnea Gets a Drug—Now Who Benefits Most?
- Post-hoc analyses of the SURMOUNT-OSA trial examined which patients responded best to tirzepatide for obstructive sleep apnea. Tirzepatide reduced breathing disruptions and hypoxic burden across subgroups defined by age, sex, BMI, and neck circumference.
- Tirzepatide became the first FDA-approved drug for moderate-to-severe sleep apnea in people with obesity in December 2024.
🧠 Higher Semaglutide Dose, Fewer Neuropsychiatric Events
- In 63,215 patients with pre-existing neuropsychiatric conditions, higher attained semaglutide doses were associated with lower rates of mood disorders, substance-related disorders, anxiety, and several other neuropsychiatric diagnoses over two years.
- The effect appeared largely independent of weight loss—except for cognitive and speech symptoms, which tracked more closely with how much weight patients lost.
👶 GLP-1 Prescriptions for Kids Ages 8–11 Are Rising
- A new study tracked GLP-1 receptor agonist prescriptions for children aged 8 to 11 from 2019 to 2026 and found a significant upward trend, particularly in children with more severe obesity and greater comorbidity burden.
- Clinical guidelines allow consideration of these drugs in this age group, but long-term safety data in young children remain limited.
Implications
GLP-1 drugs now touch oncology, neurology, aging biology, and pediatrics—all in a single week's literature. The central unresolved tension: nearly every new benefit signal comes from observational data or animal models, while the question of who should stop, switch, or never start these drugs remains genuinely open.
