APOE4 changes fat droplet proteins and alters immune responses in brain support cells

Updated

Abstract

E4 variant of apolipoprotein E (ApoE) is associated with increased lipid droplet accumulation in microglia.

  • E4 lipid droplets show a shift in glycerophospholipid distribution with more phosphatidylcholine species.
  • Baseline lipid profiles of E4 lipid droplets resemble those of LPS-treated lipid droplets.
  • E4 lipid droplets contain more proteins related to vesicle transport but fewer proteins involved in fatty acid breakdown.
  • E4 microglia accumulate more lipid droplets and secrete higher levels of proinflammatory cytokines compared to E3 microglia.
  • The LPS response in E4 microglia is reduced, indicating altered inflammatory dynamics.
