Low-cost device using aptamer-CRISPR method for detecting multiple small molecules at once

Updated

Abstract

CRISPR/Cas12a-based diagnostics achieved attomolar sensitivity for nucleic acid detection.

  • A Paper-integrated Aptamer-Cas12a Sensing platform (PACSense) enables detection of small molecules without the need for laboratory infrastructure.
  • The platform features a signal-OFF detection channel for adenosine 5'-triphosphate (ATP) with limits of detection of 336 nM in solution and 446 nM on paper.
  • A signal-ON detection channel for cocaine shows limits of detection of 0.605 µM in solution and 16.15 µM on paper.
  • Both detection channels can operate simultaneously on a single multiplexed paper chip, allowing for independent and simultaneous analysis.
  • Air-dried chips maintained 76% of initial fluorescence signal after 30 days at ambient temperature, indicating stability without cold storage.
  • The complete assay workflow takes 20 minutes and utilizes a reusable sodium acetate hand warmer and a smartphone-compatible reader.
