AsCas12a can work even when extra DNA is inserted into its target

Abstract

Essence

AsCas12a uniquely tolerates insertions in target DNA while retaining cleavage activity under some conditions.

Evidence

This in vitro CRISPR enzyme study tested 12 Cas12a orthologs, insertion lengths of 1-20 nucleotides, ssDNA and dsDNA targets, structural analysis, and cell-based activity.

Caveat

The insertion tolerance was ortholog-specific, limited on dsDNA by insertion length, position, and topology, and strongly diminished in cells.
