Asrij/OCIAD1 levels identify different types of blood-forming stem cells in bone marrow

Updated

Abstract

Asrij expression is heterogeneously present in 47% of the hematopoietic stem cell population.

  • Depletion of Asrij in mouse bone marrow hematopoietic stem cells leads to loss of quiescence and a bias towards myeloid differentiation.
  • Hematopoietic stem cells with high Asrij levels demonstrate enhanced self-renewal capacity.
  • Hematopoietic stem cells with low Asrij levels are primed for differentiation.
  • Transplantation of Asrij-expressing cells results in improved reconstitution of hematopoietic stem and progenitor cells and myeloid cells in the bone marrow.
  • Transcriptomic analysis shows that high Asrij levels are associated with signatures of quiescence, while low levels correlate with hallmarks of activation.
