Astragaloside IV may protect kidney filtering cells from damage by reducing oxidative stress through activating the PPARγ-Klotho-FoxO1 pathway in diabetic kidney disease

Updated

Abstract

Astragaloside IV (AS-IV) improved renal function and podocyte health in diabetic db/db mice.

  • AS-IV enhanced Klotho expression in glomerular podocytes, which is associated with reduced podocyte injury.
  • In vitro, AS-IV inhibited high glucose-induced podocyte apoptosis and restored Klotho levels that were diminished by high glucose.
  • Klotho knockdown reversed the protective effects of AS-IV against podocyte apoptosis.
  • Adenovirus-mediated Klotho overexpression increased antioxidant activity and reduced oxidative stress linked to podocyte apoptosis.
  • AS-IV mitigated high glucose-induced inhibition of Forkhead transcription factor O1 (FoxO1) and oxidative stress.
  • Inhibition of PPARγ blocked the effects of AS-IV on Klotho expression and its anti-apoptotic actions.
