Astragaloside IV reduces kidney cell damage in diabetic kidney disease by controlling the IRE-1α/NF-κB/NLRP3 inflammation pathway

Updated

Abstract

AS-IV treatment improved podocyte morphology and reduced markers of injury in a diabetic nephropathy model.

  • Cytoplasmic vacuolation and endoplasmic reticulum swelling were observed in high glucose conditions.
  • Treatment with AS-IV and an IRE-1α inhibitor improved podocyte morphology compared to high glucose conditions.
  • Increased expressions of GRP78 and IRE-1α were found in the high glucose group, but decreased levels were seen with AS-IV treatment.
  • Molecular markers of inflammation and cell death were elevated in the high glucose group and decreased in the AS-IV and IRE-1α inhibitor groups.
  • Diabetic nephropathy was associated with higher levels of blood glucose, triglycerides, and urinary protein, which were reduced by AS-IV treatment.
  • Pathological changes in renal tissue, including glomerular hypertrophy, were alleviated by AS-IV and IRE-1α inhibitor treatments.
