Nanomedicines targeting intestinal receptors improve semaglutide’s blood sugar control in type 2 diabetic mice

Abstract

FcRn-targeted nanoparticles improved the glucoregulatory effect of semaglutide in a type 2 diabetes model.

  • Oral delivery of semaglutide faces challenges due to limited bioavailability.
  • Nanoparticles were designed to bind to the neonatal Fc receptor (FcRn) for enhanced delivery.
  • FcRn-targeted semaglutide nanoparticles showed better blood glucose control compared to free semaglutide or nontargeted nanoparticles.
  • A similar glucose-lowering effect was observed between FcRn-targeted nanoparticles and subcutaneously administered semaglutide.
  • Increased insulin content and recovery of β cell mass were noted in mice treated with FcRn-targeted nanoparticles.
  • The nanoparticles demonstrated longer retention in the ileum and colorectum, areas with higher FcRn expression.
