Abstract

The bone-targeting mRNA nanocarrier (SA@LNP-D) significantly reduces hepatic sequestration and specifically accumulates in bones.

  • Bone-targeting peptides enhance the affinity of lipid nanoparticles for hydroxyapatite and isolated bone.
  • In vitro results show that the modified lipid nanoparticles effectively express anti-sclerostin antibody mRNA in bone tissue.
  • In a murine model of osteoporosis, the bone-targeted delivery system stimulates bone formation and inhibits bone resorption.
  • This approach significantly restores trabecular bone mass and microstructure compared to conventional lipid nanoparticles.
  • A targeted delivery strategy may provide a safe and effective therapeutic option for osteoporosis and related skeletal conditions.
