Abstract

Knockout of GIP receptors in the hypothalamus enhances weight loss effects from GLP-1 receptor agonism.

  • Distinct brain regions mediate the appetite-suppressing effects of GIP receptor agonists and the enhanced weight loss from GIP receptor antagonists.
  • Mice lacking GIP receptors in the area postrema show reduced appetite suppression and partial protection against diet-induced obesity but do not affect weight loss from GLP-1 receptor agonism.
  • Mice lacking GIP receptors in the hypothalamus maintain normal appetite suppression but exhibit greater weight loss when treated with GLP-1 receptor agonist liraglutide.
  • The synergistic weight loss effect of GIP receptor antagonists combined with GLP-1 receptor agonism is abolished in hypothalamic GIP receptor knockout mice.
  • GIP receptor antagonism and hypothalamic knockout may increase sensitivity to weight loss induced by other treatments.
