Using a Dihydropyrimidinone-Piperazine Compound to Trigger Cancer Cell Death by Targeting Growth Signals and Cell Changes in Breast Tumors

Abstract

MCB-04 demonstrated an IC50 of 20 µM against MDA-MB-231 breast cancer cells while showing minimal toxicity to normal MCF-10A cells.

  • MCB-04 exhibited potent and selective cytotoxicity towards multiple breast cancer cell lines.
  • Treatment with MCB-04 significantly increased levels of reactive oxygen species (ROS) and disrupted mitochondrial function.
  • Apoptosis was induced by MCB-04 through activation of key cell death markers such as cleaved PARP and caspase-3.
  • Increased autophagy was observed with elevated levels of LC3-II and expression of Atg5 and Beclin-1.
  • MCB-04 also indicated the involvement of paraptosis, as evidenced by increased ATF4 and CHOP markers.
  • Inhibition of c-Met signaling and downstream pathways was associated with MCB-04 treatment.
