Abstract

Exendin-4 (Ex4) increased action potential duration by 14% in isolated rat hearts.

  • Systemic administration of Ex4 increased heart rate, which was blocked by β-adrenoceptor blockade.
  • In vivo, Ex4 increased action potential duration at 90% repolarization by 7% during ventricular pacing.
  • Ex4 also reduced the inducibility of ventricular arrhythmias under β-adrenoceptor stimulation.
  • The effects of Ex4 on action potential duration and arrhythmia susceptibility were inhibited by muscarinic receptor blockade or nitric oxide synthase inhibition.
  • GLP-1R activation's impact on cardiac activity appears to involve stimulation of parasympathetic neurons.
