How DNA Shape Affects Cas9 Binding to Small Circular DNA

Closed-ring DNA shape can restrict Cas9 R-loop propagation and interfere with cleavage.

Evidence

Cryo-EM structural analysis solved two SpyCas9 complexes bound to a cognate target in a relaxed 95-base-pair double-stranded DNA minicircle.

Caveat

The evidence comes from a minicircle topology with R-loop extension limited to less than three base-pairs, so it does not directly measure genome editing across broader DNA contexts.
