Portable soil testing for detecting Phytophthora infestans using a CRISPR-based fluorescent strip and microfluidic DNA preparation

Updated

Abstract

The centrifugal Soil-on-a-Disc (SoaD) microfluidic chip reduced soil turbidity from 12,980 NTU to below detection limits.

  • The SoaD chip automates soil-to-DNA preparation, yielding clarified samples for DNA extraction in just 20 minutes.
  • Total DNA recovery efficiency for P. infestans-spiked soils was 94.9 ± 3.3% when compared to a commercial soil DNA kit.
  • The system demonstrated a limit of detection of 0.135 ng/µL for specific RPA products and identified P. infestans in soils at concentrations as low as 1 spore/g.
  • Chip-prepared DNA produced bacterial and fungal community profiles that closely matched those from a commercial DNA kit, with correlation coefficients of r = 0.996/0.923.
