Programmable Cas9 System with AI for Detecting Two Proteins and Classifying Tumor Small Vesicles

Updated

Abstract

CHAI-sEV achieved detection limits of 162 and 328 particles/µL for EGFR and PD-L1, respectively.

  • A programmable Cas9-mediated trigger-release strategy was developed for dual-protein profiling of tumor-derived small extracellular vesicles (TsEVs).
  • This method uses PAM-bearing hairpin-locking probes (PHLPs) to integrate Cas9 recognition and signal conversion into a single molecular substrate.
  • The approach enables amplification of protein-specific signals on the vesicle surface through a mechanism that avoids nonspecific collateral cleavage.
  • In plasma samples, this technique differentiated lung cancer patients from those with benign lung diseases and healthy controls.
  • Post-treatment samples showed decreased signals for EGFR and PD-L1, indicating potential for monitoring treatment response.
