Positively Charged Tiny Particles for Targeted Gene Delivery to Cartilage in Osteoarthritis Treatment

Updated

Abstract

Cationic exosomes successfully penetrated early-stage arthritic human cartilage and delivered mRNA to chondrocytes.

  • Engineered cationic exosomes were modified to enhance their ability to target cartilage for gene therapy.
  • These exosomes used a charge-reversal mechanism to interact with the negatively charged components of cartilage.
  • Intra-articular injection of mRNA-loaded cationic exosomes in mice with early-stage osteoarthritis demonstrated effective cartilage penetration.
  • The delivery system created an intra-tissue depot that efficiently expressed eGFP mRNA in deep cartilage layers.
  • Unmodified anionic exosomes were unable to penetrate cartilage or deliver mRNA effectively.
