Improved RNA detection using CRISPR/Cas12a with chemical additives

Abstract

The chemical additives-enhanced CRISPR/Cas12a-based RNA detection (CARD) achieves femtomolar-level RNA detection using only a single crRNA.

  • CARD does not require DNA activators, reverse transcription, or strand-displacement reactions.
  • This method can also be adapted for single-stranded DNA detection at attomolar levels.
  • The approach may provide a straightforward and amplification-free framework for highly sensitive diagnostics.
  • CARD is associated with significant improvements in sensitivity for RNA analysis compared to traditional methods.
