Chlorpromazine and Promethazine Reduce Excess Sugar Breakdown by Controlling HIF-1α in Stroke

C + P reduced protein levels of HIF-1α, glycolytic enzymes, and reactive oxygen species (ROS) after ischemic stroke.

  • C + P increased ATP levels in the brain following ischemic stroke.
  • In ischemic rats treated with GLUT-1/3 inhibitors, reductions in ROS, cell death, glucose, and lactate were observed.
  • C + P decreased the activation of the enzyme NOX and the expression of its subunit gp91.
  • C + P treatment improved cell viability under oxygen-glucose deprivation conditions.
  • HIF-1α overexpression diminished the effectiveness of C + P in reducing certain glycolytic markers.
