Daily rhythm gene per2 influences nerve regrowth in zebrafish escape neurons through enzyme carboxypeptidase E

Abstract

Significant inhibition of CNS axon regeneration was observed in zebrafish larvae with disrupted circadian rhythms.

  • Disruption of the circadian clock gene per2 in zebrafish larvae severely impaired axon regeneration and escape movements.
  • Time-of-day-dependent analysis showed that per2 regulates genes linked to mitochondrial metabolism and Wnt signaling, which are important for regeneration.
  • The neurotrophic factor carboxypeptidase E (Cpe) showed robust circadian rhythmicity in wild-type zebrafish but was downregulated in per2 -/- mutants.
  • Cpe deficiency significantly suppressed axonal regeneration, while its overexpression enhanced regenerative capacity in per2 -/- mutants.
