DNA-based sensors speed up Cas12a cutting to quickly detect multiple gene methylation

Abstract

A limit of detection of 74.5 aM was achieved using a novel cleavage-responsive DNA-templated silver nanocluster reporter.

  • The DNA/AgNC reporter improves detection by translating Cas12a enzyme activity into a measurable fluorescence change.
  • This reporter showed an approximately two-fold increase in catalytic efficiency compared to traditional fluorophore-quencher systems.
  • The assay's reporting step was completed within 30 minutes, enhancing the speed of detection.
  • Spatially resolved profiling of five genomic loci was achieved with relative errors of approximately 5%.
  • The approach may provide a new method for CRISPR-based DNA methylation profiling and point-of-care biosensing.
