Clinical factors linked to ketamine and esketamine antidepressant effects in hard-to-treat unipolar and bipolar depression

Abstract

A total of 122 studies involving 12,674 participants were reviewed to identify predictors of response to ketamine and esketamine in treatment-resistant depression.

  • The review included 75 distinct samples, with 65 studies administering open-label ketamine and 10 randomized controlled trials.
  • Most studies focused on treatment-resistant unipolar and bipolar depression, with intravenous ketamine typically given at a dosage of 0.5 mg/kg.
  • Of the 77 predictor variables examined, the majority did not show a significant association with treatment outcomes.
  • Promising predictive factors identified include early response to treatment and a family history of substance use disorders.
  • Further exploration of these promising factors is warranted based on the findings.
