How codon choices in modified mRNA lower immune reactions but keep vaccine effectiveness

Updated

Abstract

Et1-ethylpseudouridine (Et1Ψ) modified mRNA demonstrated comparable protein expression to N1-methylpseudouridine (m1Ψ) after codon optimization.

  • Et1Ψ-modified mRNA showed distinct codon-dependent translational sensitivity, where higher UUU codon content led to decreased protein expression.
  • Eliminating UUU codons through synonymous codon optimization restored translation efficiency to levels similar to m1Ψ-modified mRNA across various cell types.
  • Et1Ψ consistently resulted in reduced innate immune activation both in vitro and in vivo.
  • When used for vaccine antigens, Et1Ψ-modified mRNA generated strong humoral and cellular immune responses, with neutralizing activities comparable to m1Ψ-modified mRNA.
  • These results suggest that Et1Ψ is a viable nucleoside modification that can minimize innate immunogenicity while maintaining protein expression efficiency.
