Abstract

The long-term health effects of glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP) remain unclear.

  • There is controversy regarding the development of GIP agonists versus antagonists.
  • The potential effects and side effects of GIP therapies in clinical settings are not well understood.
  • Further research is needed to explore the impacts of lowering GIP alone or enhancing GLP-1.
  • Tree-structured phenotypic modeling and factorial Mendelian randomization analyses could be useful in investigating these effects.
  • Higher circulating GLP-1 levels may only indicate minimal activation achieved with DPP-4 inhibitors, not with GLP-1 receptor agonists.
