Designing Switchable Guide RNAs to Control Gene Activity

Abstract

CRISPR interference (CRISPRi) can be enhanced by using conditional guide RNAs (cgRNAs) that require specific trigger RNAs to activate their gene-silencing function.

  • cgRNAs are designed to fold into hairpin structures that prevent their activity until triggered.
  • The introduction of specific trigger RNAs allows cgRNAs to unfold and initiate transcriptional repression.
  • Activated cgRNAs can achieve large dynamic ranges and minimal unintended interactions with other processes.
  • cgRNAs offer expanded tunability and can be integrated into existing gene circuits for complex regulation of gene expression.
  • This approach provides a method for sophisticated and logical control over gene expression in living cells.
