Improving Nicotinamide Mononucleotide Production by Engineering CpNampt Enzyme Mutants in Yeast

Updated

Abstract

The I229A mutant strain achieved a total yield of 60.64 mg/L NMN in 12 hours.

  • A double-knockout mutant strain, DHSX02, was generated to enhance NMN production.
  • The introduction of a mutated NMN phosphoribosyltransferase (CpNampt) significantly increased NMN synthesis.
  • NMN yield in the DHSX02 strain increased by 20.7% compared to the wild-type strain.
  • After optimization, the I229A mutant exhibited a 4.4-fold increase in NMN production compared to the wild-type strain.
