Highly sensitive detection of methylated DNA using gene-editing enzymes combined with organic electrochemical sensors

Abstract

The platform achieved quantitative methylated DNA detection from 100 fM to 100 pM with a sensitivity of 267.6 µA/dec.

  • Methylation-sensitive detection is facilitated by AciI selectively digesting unmethylated DNA.
  • Intact methylated DNA activates the Cas12a system, which cleaves ssDNA reporters on the electrode.
  • Vertical organic electrochemical transistors (vOECTs) enhance detection through electric-double-layer gating.
  • The sensor demonstrated good operational stability and reproducibility in artificial serum samples.
  • This approach may enable sensitive epigenetic analysis and non-invasive screening strategies.
