Pregnancy Test Strip Biosensors Using CRISPR to Visually Detect Telomerase and miRNA let-7a for Cervical Cancer Diagnosis

Updated

Abstract

A detection limit of 18 HeLa cells was achieved for telomerase activity using a novel biosensor.

  • Telomerase is a key driver of cellular immortalization and is important for cervical cancer diagnosis.
  • MicroRNA let-7a acts as a tumor suppressor and is downregulated in cervical cancer.
  • Two CRISPR-Cas12a-integrated biosensors were developed to enable visual detection using pregnancy test strips.
  • The first biosensor detects telomerase activity without PCR, achieving a detection limit of 18 HeLa cells.
  • The second biosensor detects let-7a with a detection limit of 25.1 fM.
  • Clinical validation showed concordance with established methods for both biomarkers.
