Fast detection of Cardamom Mosaic Virus group IV in raw plant samples using CRISPR-based test

Abstract

The developed assay can detect Cardamom mosaic virus (CdMV) at a sensitivity level of 1 fg.

  • High genetic variability among CdMV isolates exists, approaching species-level divergence.
  • A CRISPR-Cas12a-coupled reverse transcription-recombinase polymerase amplification-lateral flow assay (RT-RPA-CRISPR-LFA) was created for detecting specific isolates from Idukki region.
  • The assay demonstrated specificity for Idukki region strains of CdMV, differentiating them from divergent isolates.
  • An incubation time of 10 minutes for RT-RPA and 1 hour for CRISPR-Cas12a reaction were optimal for performance.
  • The assay's detection limit was found to be 10 times less sensitive compared to qPCR.
