Precise DNA base editing using CRISPR in the fungus Trichoderma koningiopsis

Updated

Abstract

The mini-SDD7-CBE system achieved a maximum editing efficiency of 80% in modifying genes in Trichoderma koningiopsis.

  • Base editors, like rAPOBEC1-CBE and mini-SDD7-CBE, are designed to modify genes without causing double-strand breaks.
  • The editing window for the mini-SDD7-CBE system is primarily located at positions 16 to 20 upstream of the PAM sequence.
  • Simultaneous editing of two and three genes was successfully accomplished using the mini-SDD7-CBE system.
  • The overall editing efficiency is affected by the lowest efficiency of individual single guide RNAs (sgRNAs).
  • Application of base editing in Trichoderma is currently limited, highlighting the need for further development.
