STEPP1 is an important host protein for Epstein-Barr virus membrane protein movement and signaling, found using a gene-editing screen

Updated

Abstract

A human genome-wide CRISPR-Cas9 screen identified STEEP1 as a key factor for LMP1 trafficking in B cells.

  • LMP1, an oncogene from Epstein-Barr virus, is essential for the transformation of B cells into continuously growing lymphoblastoid cell lines.
  • STEEP1 supports the movement of LMP1 from the endoplasmic reticulum to signaling sites, which is crucial for its function.
  • Depletion of STEEP1 significantly weakened LMP1 signaling, affecting important growth and survival pathways like NF-kB and MAP kinase.
  • STEEP1 did not influence CD40 signaling, indicating its specific role in LMP1-mediated processes.
  • The findings suggest that targeting the interaction between LMP1 and STEEP1 could be a new therapeutic approach.
